β-Propeller lectins are a group of carbohydrate binding proteins consisting of repetitive protomers arranged in a propeller configuration. These lectins emerged as new targets for antimicrobial drugs due to their important roles in host cell invasion in many drug-resistant bacteria. In this work, we used NMR fragment screening coupled with computational pocket prediction algorithm to uncover compounds that target newly identified secondary pockets in a b-Propeller lectin from Burkholderia ambifaria. Our findings revealed novel non-carbohydrate binding pockets in a b-propeller lectin and a new class of compounds, which create exciting opportunities for future development of drugs combating antibiotic-resistant pathogens.
